Bloomington Vet Joins Study For Stem Cell Therapy To Treat Dogs With Arthritis – WGLT News

The Eastland Companion Animal Hospital in Bloomington is asking dog owners if they want to participate in research on using stem cells to treat dogs with arthritis.
Local dogs wouldjoin a double-blind, placebo-controlled studyto show the effectiveness of stem cells in treating large dogs(70 pounds or more) with arthritis in up to two joints of the knee, hip, elbow, or shoulder. The veterinary clinic has partnered with Animal Cell Therapies, who it's worked with before, to bring this study to Bloomington.
Dr. Kathy Petrucci, founder and CEO of Animal Cell Therapies, explained how dogs will receive the treatment.
The dogs that will receive the stem cells will be sedated, Petrucci said. Depending on what joints are affected, they will receive up to two injections in the joint and they will also receive an IV dose of stem cells.
The FDA oversees the cells that are received from donors for the study. Mothers donating these cells are screened for diseases, and cells are tested for any infections to ensure safety.
Stem cell therapy has been controversial, especially related to humans.
I think a lot of the controversy comes from the misunderstanding of the cell types, Petrucci said. The research in stem cells first started centered around embryonic or fetal tissue use. Its controversial to use embryos and fetal tissues for treatment for anything. The fact that we are using a disposable tissue as our cell sources makes it not controversial at all.
Why Umbilical-Derived Cells
Petrucci explained why umbilical-derived cells are more effective in treating arthritis versus other sources.
We looked at fat, bone marrow, embryonic cells, Petrucci said. The embryonic cells are a lot more unpredictable, and the bone marrow cells are more difficult to work with and less predictable. We didnt think the fat cells are as potent as umbilical-derived cells. Umbilical-derived cells are a lot younger and theyre a little bit more predictable. They are more easy to collect. We obtain cells from donors when the tissue would be normally thrown away. Theres no surgery required, no extra biopsies to obtain fat, no bone marrow from research animals. Its a good, ethical source of stem cells.
Umbilical-derived stem cells have proven successful in past studies on treatment for arthritis, according to Petrucci.
We did a study at the University of Florida on elbows only and we had success with that study, Petrucci said. We had good success with dogs under 70 pounds and (less) success with dogs over 70 pounds, so we changed our dose, which is why were testing dogs 70 pounds and over in this study.
Criteria for eligibility includes dogs weighing 70 pounds or more, being one year of age or older, in general good health, no neurologic issues, arthritis in up to two joints of the knee, hip, elbow, or shoulder, and have all four functioning limbs.
Owners must bring their dogs back to the clinic after 30 days to check for progress and complete a questionnaire. About 50 to 100 dogs are expected to participate in the study.

Brooks Koepka Withdraws in Korea After Reaggravating Knee Injury – Golf Channel

Brooks Koepka withdrew before the start of the third round of the CJ Cup because of a knee injury.
This fall the world No. 1 underwent a stem-cell treatment to repair a partially torn patella tendon in his left knee. On Wednesday, Koepka told reporters that his knee feels good, but he said in a statement released through the PGA Tour communications department that he reaggravated the injury when he slipped on wet concrete during a second-round 75 Friday in Korea.
Koepka was supposed to play in next weeks inaugural Zozo Championship in Japan, but now, after consulting with his doctor, hes headed home to South Florida for further testing. He was the defending champion at the CJ Cup.

Koepka never revealed his knee issue until earlier this month in Las Vegas. He said that his knee had been bothering him since March, and that it limited his practice sessions and caused him pain every time he leaned down to grab the ball out of the cup.

But other than that, I feel great. My knee feels good. Its the first time its been feeling consistently good in a while.

A man who received chimeric antigen receptor-T cell therapy shares his firsthand account.
Backer received a diagnosis of diffuse large B-cell lymphoma (DLBCL). It is the most common type of non-Hodgkin lymphoma, which affects nearly 75,000 people mostly men in the United States each year.
Despite multiple rounds of chemotherapy and a stem cell transplant, Backers disease kept relapsing. Then he participated in a clinical trial involving chimeric antigen receptor (CAR)-T cell therapy, which has left him cancer-free for almost three years. With this type of immunotherapy, a patients T cells are removed, altered in a lab and then reinfused in the hope that they will attack cancer cells.
Dr. Frederick L. Locke, a medical oncologist at Moffitt Cancer Center in Tampa, Florida, isco-lead investigator of the pivotal ZUMA-1 trial, which led to the Food and Drug Administration (FDA) approval of the second available CAR-T cell therapy, Yescarta (axicabtagene ciloleucel). During the National Comprehensive Cancer Network 2019 Annual Conference, Locke, Backer and Alix Beaupierre, a transplant nurse coordinator, took a 360-degree look at CAR-T cell therapy.
LIMITED OPTIONSOutcomes in refractory aggressive non-Hodgkin lymphoma are poor, Locke explained. Patientsare often treated upfront with combination chemotherapy, as Backer was. He initially wenton a chemotherapy regimen commonly known as R-CHOP Rituxan (rituximab), cyclophospha- mide, Adriamycin (doxorubicin), Oncovin (vincris- tine) and prednisone. He achieved a complete remission and went back to work.
We can cure up to about 60% of patients with initial chemotherapy, and thats pretty remarkable, Locke said. Unfortunately, 40% of patients either dont respond to chemotherapy or progress.
Backer was among that 40%. He relapsed about a year later and started on a new chemotherapy regimen with a planned autologous stem cell transplant, which would involve removing his own stem cells and later putting them back into his body to help fight the cancer. This treatment plan cures only about 5% of patients, Locke said. Prior to CAR-T cell therapy, more chemotherapy would have been next.
In the United States, two CAR-T therapies are available to patients with certain types of cancer. The first, Kymriah (tisagenlecleucel), was approved in August 2017 for patients up to 25 years old who have acute lymphoblastic leukemia that relapsed or did not go into remission with other treatments. Two months later, the FDA approved Yescarta to treat adult patients with certain types of large B-cell lymphoma who have not responded to or relapsed after at least two other kinds of treatment.In patients with DLBCL, durable responses the length of time that a partial or complete response is observed because of treatment have been seen in 40% of patients who received CAR-T cell therapy. We think we can cure about 15% of people, Locke said. We need these patients referred and referred early.
A NUCLEAR BOMBBacker first read about CAR-T cell therapy on clinicaltrials.gov, an online registry of all clinical trials that anyone can access to see what might be enrolling participants. Im the typeof person who needs a plan A and a plan B, Backer said.I received a plan A, but the plan B in the case of the transplant failing was not encouraging.
He reached out to Moffitt Cancer Center Tampa is not far from where he lived in Orlando, Florida to see if he was eligible. He wasnt. In December 2015, Backer went ahead with the stem cell transplant. A few months later,he again relapsed with growths all over his body butthis made his participation in the ZUMA-1 clinical trial possible. Being a participant in a clinical trial is scary and daunting at the same time, he said. I remember sitting there with the transplant coordinator and they handed me a 27-page consent form, and I could barely read page one. Ijust wanted to sign. I was ready to sign anything right then and there.
Although potentially curative, CAR-T cell therapy is not without risk. Patients can develop two serious side effects. Cytokine release syndrome, caused by a large, rapid release of cytokines (small proteins importantin cell signaling) into the blood from immune cells affected by the immunotherapy, can be life-threatening. Neurological events, such as confusion, tremor and seizures, can also occur.
Despite lengthy discussions with his medical teamat Moffitt and reading the consent form, Backer said,he wasnt fully prepared for the coming side effects and recovery when he went forward with CAR-T therapy in June 2016. Thats when the nuclear bomb set off, he said. Within 12 hours of receiving the infusion, he experienced severe chills, violent shaking and a high fever, and also felt certain he was experiencing side effects that were affecting his brain.
About two days after Backer received the reinfusedT cells, the infectious disease team rushed him in for a CT scan. They found no infection and no signs of the cancer.
For me, it was a miracle treatment, Backer said.
Locke has been following patients enrolled in ZUMA-1 for more than two years and said that half are still alive.
BEING PREPAREDBacker admitted that going in with a full understanding of CAR-T cell therapy would have made the experience dramatically different.
After seeing patients and their loved ones in distress, care providers at Moffitt Cancer Center learned to smooth the process, Beaupierre said. For instance, the 27-page document has been broken down into one-page educational handouts and shorter consent forms. The team also created a flow sheet and that grew to a detailed patient calendar. All CAR-T therapy recipients also now have a dedicated nurse and social worker.
In addition to those resources, Backer said, peer-to-peer support would be helpful.
As CAR-T cell therapy continues to evolve and be explored in the treatment of other cancer types, experts are learning more about how it works and how to improve the process.
For Backer, quality of life has changed a bit. Initially, he received blood and platelet transfusions every two weeks for several months following CAR-T cell therapy. Although he is back to work full time, he runs the risk of being exposed to other diseases and viruses. Its always at the back of my mind, he said. I wear a mask and goggles at work but still get sick.
He spends his free time hiking and fishing and feels blessed to still be alive. It worked out for me, and here we are 33 months into this thing, Backer said.

The Real Problem With Beef – The New York Times

The potentially unhealthful effects of eating red meat are so small that they may be of little clinical significance for many people.
This finding, just released in multiple articles in the Annals of Internal Medicine, is sure to be controversial. It should certainly not be interpreted as license to eat as much meat as you like. But the scope of the work is expansive, and it confirms prior work that the evidence against meat isnt nearly as solid as many seem to believe. (While I had no role in the new research, I co-wrote a commentary about it in the journal.)
Red meat has been vilified more than almost any other food, yet studies have shown that while moderation is important, meat can certainly be part of a healthy diet.
This doesnt mean that there arent other reasons to eat less meat. Some point out that the ways in which cattle are raised and consumed are unethical. Others argue that eating red meat is terrible for the environment.
Recently, meat substitutes have emerged as a possible solution, but the promise is much greater than the reality, at least so far.
Burger King and other fast-food chains are trying out Impossible Foods burgers as a vegan answer to beef. Lets dispense with the idea that this is healthier in any way. The Impossible Whopper has 630 calories (versus a traditional Whoppers 660). It also contains similar amounts of saturated fat and protein, and more sodium and carbohydrates. No one should think theyre improving their health by making the switch.
What about the environmental argument? Almost 30 percent of the worlds ice-free land is used to raise livestock. We grow a lot of crops to feed animals, and we cut down a lot of forests to do that. But beef, far more than pork or chicken, contributes to environmental harm, in part because it requires much more land. The greenhouse gas production per serving of chicken or pork is about 20 percent that of a serving of beef.
Cows also put out an enormous amount of methane, causing almost 10 percent of anthropogenic greenhouse gas emissions and contributing to climate change.
There has been a lot of hope that Beyond Meats pea protein or Impossible Burgers soy could serve as beef burger substitutes, reducing the need for cows. Thats unlikely to happen, according to Sarah Taber, a crop scientist and food system specialist. Ground beef is not the problem; steak is.
Theres no profit to be made in ground beef, she said. That all comes either from leftover parts once cattle have been slaughtered for more expensive cuts, or from dairy cattle that have outlived their usefulness. If everyone gave up hamburgers tomorrow, the same number of cows would still be raised and need to be fed.
In other words, to improve the environment by reducing the number of cows slaughtered, wed need to find a way to replace the many other cuts of beef Americans enjoy. No lab, and no company, is close to that.
To greatly reduce the reliance on cows, wed also need to wean ourselves from our high level of milk consumption. The increasing use of alternative milks, like oats or soy, could help, but the dairy industry still dominates.
(The dairy industrys claims about the health of its product are somewhat overblown. Milk isnt nearly as necessary for health as many believe.)
Some companies are researching ways to replace the more complex cuts of meat that drive the market. These companies arent replacing beef with substitutes; theyre trying to grow it in the lab using stem cells.
Tamar Haspel, who writes on food policy for The Washington Post, has said such advances are not likely soon. Nor is it clear that they would have an overall positive impact, unless we are sure that this meat can be made in a more energy-efficient way than we can raise cattle.
If meat substitutes wont help in the short run, other things still might. Some believe that raising cattle on pastures, from birth until slaughter, might sequester carbon in the soil better than having cows finish their growth on feed lots. Researchers at the University of Florida argue that it can also be profitable for farmers in warmer climates to do just that. It would require the cattle industry to make significant changes, as well as to relocate, and it seems unlikely theyd be willing to do that.
Grass-feeding cattle without grain is the norm in New Zealand, but almost no one in the United States does it, Dr. Taber said.
Its also worth pointing out that it would probably take longer to raise cows this way, giving them more time to emit methane.
Other new developments could help with that problem. Some have proposed farming insects to make animal feed. And feeding seaweed to cows, even in small amounts, can significantly reduce their methane burps.
One problem with seaweed is that the component that helps reduce methane emissions is classified as a carcinogen by the Environmental Protection Agency. Its present in small amounts in seaweed, though, and humans have been eating seaweed safely for a long time. A larger problem is that we are unprepared to farm the unbelievable amount of seaweed it would take to feed all the cattle the world is raising.
Picture a seaweed farm the size of Manhattan, Dr. Taber said.
Until people are truly ready to reduce consumption of dairy or consumption of higher-end beef cuts, or to commit to raising cattle differently, it seems unlikely that any of the changes with respect to ground beef will make a significant environmental difference in the near future.
That doesnt mean theres nothing we can do. I asked Dr. Taber what we might advise people, right now, to help the environment.
Who needs steak when theres bacon and fried chicken? she said.

What conditions can respond to stem cell therapy?
Stem cells can help heal almost any joint in the body. What is Erectile Dysfunction or ED?
Erectile Dysfunction, sometimes called 'impotence', is the repeated inability to get or keep an erection firm enough for sexual intercourse. The word 'impotence' may also be used to describe other problems that interfere with sexual intercourse and reproduction, such as lack of sexual desire and problems with ejaculation or orgasm. Using the term erectile dysfunction makes it clear that those other problems are not involved.
Damage to nerves, arteries, smooth muscles, and fibrous tissues, often as a result of a disease, is the most common cause of erectile penile dysfunction. Diseases such as diabetes, kidney disease, chronic alcoholism, multiple sclerosis, atherosclerosis, vascular disease, and neurologic disease, account for about 70 percent of erectile dysfunction cases. Between 35 and 50 percent of men with diabetes, experience erectile dysfunction.
What causes ED?
Defects preventing an appropriate erectile tissue response can interfere with reproductive capability. Erection of the penis is a hemodynamic action that tissue of the corpora cavernosa and the corpus spongiosum. This tissue consists of smooth muscle, endothelial cells, fibroblasts, and nerves that interact in order to enhance and maintain an accessory blood supply causing erection. Given the need for the blood flow during this response, it is clear that vascular insufficiency can decrease erectile capability.
In fact, vascular insufficiency may be a common pathology of erectile dysfunction. It is estimated that up to 80% of the cases of erectile dysfunction can be attributed at least in part to a reduced blood flow, typically due to an underlying vascular disease.
Pharmacological methods have been used to treat erectile dysfunction including the use of vasodilators. Such vasodilators include Viagra and Cialis. However, vasodilators induce only a temporary response and are not effective in many patients due to the underlying endothelial damage. Not to mention the fact that these medicine can have serious side-effects including heart attack, stroke and may even cause death.

It is important to understand that lipoaspiration is vastly different from liposuction. Liposuction removes a very large volume of fat and is intended for a cosmetic result. Lipoaspiration uses a small amount of local anesthetic, a small instrument slightly larger than a hypodermic needle, removes a very small amount of fat (10 20cc), and does not render any visible change in appearance. The fat is then concentrated using a simple, sterile gravity method. The entire process is painless and takes under ten minutes.
Bone Marrow Stem Cells
Bone marrow contains stem cells; primarily hematopoietic stem cells. Bone marrow is harvested from the hip (iliac crest) using local anesthetic and a specialized needle. It is important to understand that bone marrow harvesting for BMAC is vastly different from bone marrow harvesting for transfusion for cancer therapies. In bone marrow harvesting for transfusion, a hematologist, oncologist, or surgeon removes a very large amount of bone marrow. BMAC requires only a very small amount of bone marrow. Surprisingly, the procedure is not painful, takes under ten minutes, and causes only mild soreness afterward. It has an excellent safety profile.
Platelet-Rich Plasma (PRP)
PRP is an acronym for Platelet Rich Plasma. Plasma is essentially blood without the red blood cells. Platelets are cells found in the blood that are essential for clot formation and wound healing. PRP, the mix of concentrated platelets and serum, contains factors that promote repair of tissues. PRP is injected into the targeted area for repair.
Packed with growth and healing factors, platelets initiate repair and attract the critical assistance of stem cells. PRP therapys natural healing process, by providing required growth factors, intensifies the bodys efforts to renew and rejuvenate your aging face. The Procedure:

Is it painful?Most of my patients report feeling mild pressure during the injection that is controlled with local anesthesia and some soreness in the areas treated for up to 5 to 7 days after the procedure that does not require pain medication.
How long is the recovery?You will be able to resume your normal daily life immediately, limited by the degree of swelling that you experience after the procedure. Most patients have noticeable swelling in the areas treated from 5 to 7days depending on the extent of the procedure and individual healing response.
How long will the result last?The results should last for several years. The results are affected by your lifestyle, habits and genetics. Smoking and weight fluctuations diminish the results.
Is Stem Cell Therapy Approved by FDA?FDA does not regulate medical procedures performed by qualified and licensed doctors.
