Protein-based compounds include collagen, fibrin, fibrinogen, and silk. Collagen seems to be the most popular of these. I can be transformed into a 3 dimensional matrix. Stem cells are attracted to the scaffold and form blood vessels inside it. Multiple studies have demonstrated that the use of these 3-D collagen scaffolds along with stem cell seeding may have utility in the construction of ligaments, cartilage, and bone. Fibrin, while not studied as much as collagen, also appears to have promise. Silk, used as a 3-D scaffold also has promise. Meinel and colleagues demonstrated that human mesenchymal stem cells combined with silk scaffolds produced more cartilage than comparable collagen 3D scaffolds. Silk has tensile strength that makes it attractive for engineering connective tissue. http

Corticogenesis from Pluripotent Stem Cells
In this video, Ira Espuny-Camacho and Pierre Vanderhaeghen describe how human pluripotent stem cells can be turned into cortical pyramidal neurons, mimicking key aspects of human corticogenesis. Most strikingly, they show that these human neurons can be transplanted successfully in the mouse neonatal brain, where they integrate specifically and functionally like endogenous cortical neurons. These data provide a unique framework to model human cortical development in health and disease, both in vitro and in vivo. For more information, see Espuny-Camacho et al., Neuron 77(3).

Five years ago, Czaja and WSU colleagues discovered by accident that a nerve center just outside the brain can recover from damage by inducing developing stem cells to become functional nerve cells, or neurons. Now he is looking for the substances that get the process going. "Our idea is to trigger the cells and then plant them in a new, injured environment so they start to develop and seek connections," Czaja says. The grant is one of 15 that NFL Charities awarded last year to support sports-related medical research, mostly in the prevention and treatment of concussions. Biomaterials have been defined at the European Society of Biomaterials Conference in 1987 as " non-viable materials used in a medical device, intended to interact with biological systems." What all this means as far as tissue engineering is that using a framework along with stem cells seems to be promising. The logical types of scaffold material are the natural proteins and polysaccharides (sugars) that make up normal connective tissue. These are biocompatible and contain sites where stem cells can congregate. For practical use these materials need to be able to be produced in a consistent manner and they also have to be modified so they provide sufficient mechanical structure to hold up following transplantation. http
