North-east man urges stem cell donation to save lives – Aberdeen Evening Express

An Aberdeen man who saved a life by donating his stem cells has urged others to do the same.
Paul Flecher-Herd, from Kingswells, signed up to the Anthony Nolan register in memory of his friend Cammy Smith, who passed away from leukaemia in 2014, aged just 26.
This year, Paul found he was a match with someone battling the same condition, and donated his cells during a procedure in London.
He has since heard from the recipient, thanking him for saving his life.
Now Paul, 28, has appealed for more people to join the register.
He said: I think having that first-hand experience makes it easier for people to come to me and ask what its like. A lot of people have said they wouldnt have thought of doing it because they either didnt know what it was for or because they were scared of the procedure.
But once I explained it to them, and how it can impact on the recipients life, they soon changed their mind.
Its four hours. Although there is medical prep before it, time like that is nothing if you get the result you could possibly get. You could save someones life, and if thats the case, it doesnt matter how long it takes.
Paul wants to put peoples minds at ease if they have any concerns over the procedure and he said the results of donating cells make it worthwhile.
He said: I would tell anyone to just go for it. You could save someones life and thats something you cant beat.
It may sound daunting but once you realise what the outcome may be you should definitely go for it and pass on the message to anyone.
It has potentially amazing consequences at the end of it. You could give someone a second chance at life.
This year Paul also launched a petition aimed at bringing in laws requiring employers to give staff time off to donate.
And although it was cancelled by the government as a result of dissolution of Parliament for the general election, he was thrilled with the response.
Paul added: With what we have done so far in terms of raising awareness, so many people have asked me about it.
A lot of employers have said they are keen to adapt their processes for the sake of the people who need these procedures.
They want to make sure they are not restricting their workers by not giving them time off. Its great to see the response its had.
There have been so many messages asking about it and Ive even been stopped in the street.
Its raised a lot of awareness about the issue of stem cell donation.

According to the researchers of the study, this is the first time that teeth-like structures have been regenerated in a living organism.
There can be a lot of benefits that come along with the new procedure if this treatment proves successful in humans. Since the tooth is grown in the socket where it will stay, there is also no need to harvest outside stem cells or make an outside environment for the tooth as it grows.
Because of this experiment, the researchers hope that this will be a better and more cost-effective solution for patients who can't afford dental implants. Also, since the tooth grows right in the mouth, there will likely be less recovery time and it will be less likely for teeth to fail.
However, for the time being, dental implants are the closest thing that we have to help replace lost teeth, Unfortunately, implants can be painful and it requires a long healing process, also it does not usually adapt to aging mouths which can cause the implant to fail.
Disadvantages of dental implants
The first disadvantage ofdental implantsis that they are expensive. Dental implants are not covered by dental insurance, if anything, they might help cover the restoration that will be attached to the dental implant such as the dental bridge, dental crown, and partial or full denture.
Another disadvantage of dental implants is that they need surgery to be placed. This may not be a big deal for some but keep in mind that surgery always has a health risk. The complication rate is just an average of 5 to 10%. The complications and risks of dental implants include damage to other teeth, infection, nerve damage, delayed bone healing, jaw fractures, prolonged bleeding and more.
With that said, this new alternative may become available soon. Even now, researchers at Columbia University have filedpatent applicationsfor this discovery and they are also looking at ways to make the process available commercially.

Researchers have established a baseline set of injectable hydrogels that show promise to help heal wounds, deliver drugs, and treat cancer.
Critically, theyve analyzed how the chemically distinct hydrogels provoke the bodys inflammatory responseor not.
The researchers designed the hydrogels to be injectable and create a mimic of cellular scaffolds in a desired location. They serve as placeholders while the body naturally feeds new blood vessels and cells into the scaffold, which degrades over time to leave natural tissue in its place. Hydrogels can also carry chemical or biological prompts that determine the scaffolds structure or affinity to the surrounding tissue.

The study demonstrates it should be possible to tune multidomain peptide hydrogels to produce appropriate inflammatory response for what theyre treating.
Weve been working on peptide-based hydrogels for a number of years and have produced about 100 different types, says Jeffrey Hartgerink, a chemist and bioengineer at Rice University. In this paper, we wanted to back up a bit and understand some of the fundamental ways in which they modify biological environments.
The researchers wanted to know specifically how synthetic hydrogels influence the environments inflammatory response. The two-year study offered the first opportunity to test a variety of biocompatible hydrogels for the levels of inflammatory response they trigger.
Usually, we think of inflammation as bad, Hartgerink says. Thats because inflammation is sometimes associated with pain, and nobody likes pain. But the inflammatory response is also extremely important for wound healing and in clearing infection.
We dont want zero inflammation; we want appropriate inflammation, he says. If we want to heal wounds, inflammation is good because it starts the process of rebuilding vasculature. It recruits all kinds of cells that are regenerative to that site.
The labs tested four basic hydrogel typestwo with positive charge and two negativeto see what kind of inflammation they would trigger. They discovered that positively charged hydrogels triggered a much stronger inflammatory response than negatively charged ones.
Among the positive materials, depending on the chemistry generating that charge, we can either generate a strong or a moderate inflammatory response, Hartgerink says. If youre going for wound-healing, you really want a moderate response, and we saw that in one of the four materials.
But if you want to go for a cancer treatment, the higher inflammatory response might be more effective, he says. For something like drug delivery, where inflammation is not helpful, one of the negatively charged materials might be better.
Basically, were laying the groundwork to understand how to develop materials around the inflammatory responses these materials provoke. That will give us our best chance of success.
Researchers at Texas Heart Institute (THI) helped analyze the cellular response to the hydrogels through multidimensional flow cytometry.
The results of this work lay the groundwork for specifically tailoring delivery of a therapeutic by a delivery vehicle that is functionally relevant and predictable, says Darren Woodside, vice president for research and director of the flow cytometry and imaging core at THI. Aside from delivering drugs, these hydrogels are also compatible with a variety of cell types.
One of the problems with stem cell therapies at present is that adoptively transferred cells dont necessarily stay in high numbers at the site of injection, he says. Mixing these relatively inert, negatively charged hydrogels with stem cells before injection may overcome this limitation.
Hartgerink says the work is foundational, rather than geared toward a specific application, but is important to the long-term goal of bringing synthetic hydrogels to the clinic.
We have been speculating about a lot of the things we think are good and true about this material, and we now have more of a sound mechanistic understanding of why they are, in fact, true, Hartgerink says.

Look under the hood of many of this years headline-making discoveries in biology and youll find machine learning, a tool thats gaining ground in the life sciences thanks to growing computational power and the availability of big datasets needed for training. Among other advances in 2019, researchers reported successfully using machine learning to screen images for signs of cancer or infection by pathogens, and to identify epigenetic markers in blood samples that are associated with vascular complications in people with diabetes. Even as computers take on more of the tasks once done by hand, engineers are exploring DNAs capacity to adopt a function usually associated with machines: information storage. This summer, researchers in Boston reported a way of harnessing DNA, together with CRISPR-like base editing machinery, to make a record of events inside living cells that can then be decoded via sequencing. Study coauthor Timothy Lu of MIT told The Scientist that its potential applications include detecting environmental toxins and recording developmental processes.
Another creative spin on CRISPR-Cas9 editing to come out this year is a detection device for particular DNA sequences. Here, the Cas9 enzyme is bound to an RNA and to a graphene chip and engineered not to make cuts in DNA. If the RNA-Cas9 complex connects to its target DNA sequence, it causes a change in the chips electric field and thus a positive readout. The chips developers suggest it could one day be used for quick DNA tests in clinical settings.
Among the endless variations of CRISPR scientists are engineering, one developed this year purports to reduce its off-target effects by avoiding double-strand DNA breaks. The technique, known as prime editing, uses the same Cas9 nuclease as frequently deployed in the CRISPR system but combines the enzyme with a guide RNA called pegRNA and a reverse transcriptase that initiates the addition of a new sequence or base into the genome. Once the new genetic material is incorporated into a cut strand of DNA, the prime editor nicks the unedited strand, signaling to the cell to rebuild it to match the edited strand.
As some researchers worked on their own variations of genome editing, others made an important edit of a recipe for induced pluripotent stem cells. First published by Shinya Yamanaka (now of Kyoto University) in 2006, the method overexpresses genes for four transcription factors in differentiated cells to reset them to a pluripotent state, creating what are known as induced pluripotent stem cells (iPSCs). The most important of the four overproduced factors was thought to be Oct4. But last month, researchers at the Max Planck Institute for Molecular Biomedicine announced theyd not only managed to make mouse iPSCs without tweaking Oct4 levels, but that the process was more efficient that way. If this works in adult human cells, it will be a huge advantage for the clinical applications of iPS cells, Yamanaka wrote in an email to The Scientist.

In 2010, Gwyneth Paltrows wellness brand, Goop, was just starting to get its goop-y mojo rolling. Tom Bradys lifestyle company, TB12, wasnt around, so we had no way of learning about bogus fitness concepts like muscle pliability. And Jessica Albas The Honest Company, a fearmongering and pseudoscience-based business that is currently worth over a billion dollars, was still one year away from inception.
But what a difference 10 years has made. Now all of these companies are thriving and many other celebrities, including Victoria Beckham and Kate Hudson, have started similar wellness brands.
But it is hard to deny that things are qualitatively different now. This has been the decade of misinformation. And, in the context of health, celebrities have led the charge.
Yes, pseudoscientific health claims have been with us for a long time. And celebrities have often embraced them. (Apparently, Greta Garbo never met a fad diet she didnt like or, at least, try.) But it is hard to deny that things are qualitatively different now. This has been the decade of misinformation. And, in the context of health, celebrities have led the charge.
Weve had the vagina steam (thanks, Gwyneth), jade vagina eggs (ditto), the vampire facial (Kim Kardashian West), bird poop facials (David and Victoria Beckham), facials made with discarded foreskin stem cells (Sandra Bullock), drinking your own urine (Madonna), placenta smoothies (more Kardashians) and too many crazy diets, cleanses and detoxes to mention. I could go on and on and on.
It seems entirely appropriate that we are closing this ridiculous decade with the too-absurd-to-be-true (but it is true) news that Josh Brolin burned his anus trying the latest wellness trend, perineum sunning.

Some may dismiss the critique of celebrities and the associated health fads as a waste of time. Few people take this stuff seriously, it is argued. Bigger fish to fry. Fish in a barrel.
This perspective is mistaken.
Celebrity health noise has had (and continues to have) a large and measurable impact. There is a growing body of literature that has demonstrated celebrity marketing, musing and news coverage can have an influence on a range of health related behaviors, including dieting, cancer screening, smoking and suicide. Pop culture coverage of a health topic, like Angelina Jolies decision to get genetic testing, can affect, for better or worse, the utilization rates of health services. And there seems little doubt that many current evidence-free and potentially harmful health trends such a IV vitamin therapy, nonceliac gluten-free diets, cryotherapy and detoxification diets and procedures would not be nearly as popular but for the associated celebrity endorsements.
In addition, all this celebrity noise and wellness-related pontificating adds to an already noisy health information environment. Studies have consistently found that the public is increasingly confused about what a healthy lifestyle entails this, despite the fact that for most people the essential ingredients are straightforward and well established (dont smoke, exercise, eat real food, sleep, maintain a healthy weight, and drink alcohol in moderation or not at all).
When it comes to public discourse, few entities have the volume and reach of celebrities. As I write this, Katy Perry has 108 million Twitter followers; the World Health Organization has 5 million. In 2010, Instagram was just getting started. Ten years later, Instagram has emerged as a significant source of health misinformation and much of the messaging on the platform is dominated by celebrities (currently 17 of the top 20 Instagram accounts are run by either a musician, an actor or a sports star). When Katy Perry tweets about her love of supplements or Tom Brady posts science-free diet advice, it is seen by tens of millions.
Just being around this social media-fueled celebrity health noise can have an impact on our health behaviors and beliefs. The more we hear about something, the more believable it becomes. This is how and why fake news works. Indeed, research led by Canadian psychologist Gordon Pennycook has found that even a single exposure to misinformation can affect perceptions of accuracy.
And when celebrities do provide health advice be it about the effectiveness of an extreme diet, a ridiculous waist-training device, anti-vaccine baloney, or the need to screen for prostate cancer it is often packaged in the form of a compelling story. Narratives, especially highly memorable ones, can be extremely influential. A persuasive testimonial can displace a mountain of scientific data. Indeed, a 2016 study found that anecdotal stories impede our ability to reason scientifically.

I believe this is one of the reasons why celebrities hold so much sway. Celebrity wellness gurus are not truly health experts. But their messaging still has power because it plays to our cognitive biases, including the mere-exposure effect and our hardwired tendency to be influenced by stories.
What is a celebrity endorsement, after all, but a glossy, high profile and impressive testimonial from someone who is often a genetic outlier in areas such as appearance and athletic ability? When Tom Brady recommends that we avoid the consumption of dairy, it may feel like a good idea because it seems to have worked for him. Do not be fooled. You arent Tom Brady (unless you are, in which case, enough with the diet nonsense).
Of course, this decade of celebrity health hogwash should also be considered in the broader context. This is the era of misinformation, a time when trust in public institutions is declining and people feel uncertain about what to believe about, well, everything. Celebrity wellness hype contributes to this culture of untruth by both inviting a further erosion of critical thinking and promoting what is popular and aspirational rather than what is true.
In the coming decade let's do our best to ignore the celebrity noise (a man can dream!). We need everyone who cares about accurate representations of science and health issues including researchers, public health advocates, health care institutions, universities and, hopefully, you to use creative communication strategies, engaging story telling and social media-friendly imagery to get across the good science. Lets fight the celebrity-fueled misinformation tire fire with a fact-filled fire of our own.
