What started in 2009 as an after-school program has become Future Kings, a robust non-profit organization delivering immersive science, technology, engineering, and math (STEM) programs to middle and high school boys from underserved communities through a multi-year, hands-on curriculum, and dedicated mentorship.

My vision for Future Kings and my ongoing commitment to the organization is to break the cycle of poverty in African American and Hispanic communities by introducing students to STEM professions, says Dr. King, Future Kings Founder and Executive Director.

As the organization evolved, a new biomedical sciences program emerged in collaboration with Northern Virginia Community College. It helps students build technical skills; 21st century soft skills such as adaptability and collaboration; and applied learning that puts classroom knowledge to work in a real-world environment. In 2015, Dr. Tshaka Cunningham, Co-Founder and Chief Scientific Officer at Polaris Genomics, joined the Future Kings board and became a mentor. The program expanded further to provide the tools and support for life after high school.

Our goal for the expanded program is to expose young men to cutting edge techniques in molecular and cellular biology, says Dr. Cunningham. It is a very exciting program because it provides hands-on job skills that give students a competitive advantage over their peers as they apply for college, internships and jobs in the future.

Dr. Cunningham is also a graduate of the Illumina Accelerator program associated with the team at TruGenomix, now Polaris Genomics. Polaris addresses solutions for post-traumatic stress disorder (PTSD) by offering better identification of the condition and targeted, individualized treatment options.

Armed with the knowledge of Illuminas sequencing systems and reagents, Dr. Cunningham connected the Future Kings team to Illumina in 2020 for support in building out the biomedical sciences program to include industry-leading equipment and resources that the students wouldnt have had access to otherwise.

We are committed to equitable access to STEM education opportunities through the support of programs like Future Kings, says Sharon Vidal, Global Lead of Corporate Social Responsibility at Illumina. Genomics is transforming science, and the diversity of future scientists and engineers will be critical for innovation and inclusive solutions.

In addition to students building basic lab skills, nucleic acid structure and manipulation, cell structure and function, protein structure and analysis, drug development, and entrepreneurship, they are also able to choose the projects they work on.

In the fall of 2020, the cohort of 12 students began their capstone research project. They chose to study why African Americans are more susceptible to COVID-19. Recent studies show that certain minority communities are at higher risk for fatal COVID-19 infection. In parts of the United States, the mortality rate for African Americans was six times greater than other races, for reasons that remain unknown.

For the project, students aimed to answer this questionto uncover genes that define cellular susceptibility or resilience to COVID-19 infection utilizing Illuminas sequencing technology. The hope is that cloning under-expressed genes from the resistant cells and introducing them into susceptible cells could help people become more resistant. Ultimately, the goal would be to use the research and documentation to help develop new gene-based treatments for COVID-19.

Over the course of the year, students learned how to perform sterile technique, pipetting technique, DNA/RNA extraction, gel electrophoresis, human cell culturing and library preparation using Illuminas iSeq technology and related components.

To me, Future Kings means mentorship and commitment. Having mentors with as much expertise as Dr. Cunningham and being enriched every other weekend for a few hours is a unique experience you cant get anywhere else, and you cant get that in school, says Norvin West, a Future Kings graduate who has started his freshman year at Yale where he will pursue a degree in neuroscience.

Other graduates of the program have gone on to study virology, pre-med, and mechanical engineering at Johns Hopkins, Virginia Commonwealth University, and George Mason University, respectively.

Light-Based Therapy Shows Promise in Treating Mucositis in Pediatric Cancer Patients – Cancer Therapy Advisor

A new light-based device for wound healing is showing excellent early results in the treatment of mucositis in pediatric cancer patients.1 The therapy, referred to as photobiomodulation, uses low-level lasers in the near-infrared range to stimulate and promote wound healing and regeneration. The device is being studied in the prevention and treatment of mucositis in pediatric cancer patients at West Virginia University (WVU) School of Medicine in Morgantown and other centers in the United States.

Mucositis is characterized by painful and debilitating sores in the mouth, tongue, oropharynx, and throat. It is caused by certain treatments common to cancer regimens, particularly chemotherapeutic agents that preferentially affect fast-dividing cells. Along with the direct effects on the oral cavity, therapy-associated immunosuppression can result in dramatically reduced wound healing. The mucositis generally wont resolve until the immune system recovers, which depending on the type of chemotherapy used, can result in 7 to 14 days of sores and oral pain, said Patrick Tomboc, DO, chief of the Division of Pediatric Hematology-Oncology at WVU, who is using the new light-based device on his pediatric oncology patients.

The pain caused by mucositis can sometimes be so extreme that children require intravenous pain medication and feeding tubes. Aversions to food and oral medication can develop in very young children as a consequence of enduring mucositis-associated pain. Affected children are also at risk for infection via oral sores. Some of the worst mucositis I have seen has been in the setting of bone marrow transplantation. The throat can be so inflamed that the child vomits throat tissue. Mucositis also can lead to serious infections, as the mucosal barriers are often our best way to protect against infection, said Dr Tomboc.

The photobiomodulation device is thought to improve tissue repair by reducing inflammation via reduced oxidative stress in mitochondria.2 The treatment involves inserting a probe-like device into the mouth for 1 to 2 minutes per treatment. It also can be applied on the outside of the skin in very young patients whose oral cavity is too small for the device.

Currently, the device is used prophylactically 3 times a week in patients who are at risk for mucositis or daily as a treatment in those in whom mucositis has developed. Because limitations such as device size exist in the use of photobiomodulation in the pediatric setting, Dr Tomboc and his team are developing a smaller device. It seems to be a painless intervention, and I dont know of any side effects, he explained. The team is planning a prospective study to evaluate photobiomodulation in pediatric cancer patients with mucositis.

In relating observations regarding device efficacy, Dr Tomboc noted that hospitalization time can be greatly reduced. One patient admitted for mucositis was in for 13 days and on patient-controlled analgesia for 10 days. When the same patient was admitted a second time, he was treated with photobiomodulation. Hospital stay was reduced to 4 days, and his mucositis resolved long before his white blood cell counts recovered, said Dr Tomboc. Clinically, the results have been very surprising. Although the number of patients that we have treated so far has been small, we have seen very effective results. We have achieved resolution of mucositis without immune system suppression, he added.

Low-level laser therapy is indicated by a multinational supportive care association for treatment of oral mucositis in adults, explained Geraldine Jacobson, MD, chair of the Department of Radiation Oncology at WVU. We have been using this treatment in the Department of Radiation Oncology since June to reduce oral mucositis in our head and neck cancer patients.

Studies on the benefit in children so far have been inconclusive.3 A clinical trial that is looking at photobiomodulation in pediatric patients who have received hematopoietic stem cell transplants is currently underway (NCT04227340).

Weve seen some amazing responses, said Belinda N. Mandrell, PhD, RN, principal investigator of the clinical trial and director of Division of Nursing Research at St. Jude Childrens Research Hospital in Memphis, Tennessee. We have a few patients who did not get the light therapy with first transplant but did with subsequent transplants. One patient had grade 3 mucositis before light therapy. Following light therapy with second transplant, no mucositis developed at all. The patients and their families are very impressed with the difference that light therapy has made, said Dr Mandrell.

The clinical trial has recruited approximately 60 patients, most of whom have acute lymphoblastic or acute myelogenous leukemia and are undergoing hematopoietic stem cell transplantation. In this trial, the light therapy is being used preventatively, with the results being compared with historical controls. We start the therapy on the first day of conditioning and continue until Day 20 or engraftment is confirmed for 2 consecutive days. Weve treated children as young as 4 months without difficulty, she explained.

Dr Mandrell and colleagues will be doing an interim analysis shortly, with a look at metrics of success, such as days of hospitalization, use of pain medication, and emergence of infections. Although results of the trial are pending, she has been impressed with the therapy so far and hopes that it will become more widely used. After I look at the study data, I want to get the evidence into clinical practice to help prevent oral mucositis. I will then look outside of the hematopoietic stem cell transplant population to other patients, such as those getting high-dose chemotherapy, said Dr Mandrell.

FDA Cracks Down On Stem-Cell Clinics Selling Unapproved Treatments – West Virginia Public Broadcasting

The Food and Drug Administration is cracking down on "unscrupulous" clinics selling unproven and potentially dangerous treatments involving stem cells.

Hundreds of clinics around the country have started selling stem cell therapies that supposedly use stem cells but have not been approved as safe and effective by the FDA, according to the agency.

"There are a small number of unscrupulous actors who have seized on the clinical promise of regenerative medicine, while exploiting the uncertainty, in order to make deceptive, and sometimes corrupt assurances to patients based on unproven and, in some cases, dangerously dubious products," FDA Commissioner Scott Gottlieb said in a statement Monday.

The FDA has taken action against clinics in California and Florida.

The agency sent a warning letter to the US Stem Cell Clinic of Sunrise, Fla., and its chief scientific officer, Kristin Comella, for "marketing stem cell products without FDA approval and significant deviations from current good manufacturing practice requirements."

The clinic is one of many around the country that claim to use stem cells derived from a person's own fat to treat a variety of conditions, including Parkinson's disease, amyotrophic lateral sclerosis (ALS), and lung and heart diseases, the FDA says.

The Florida clinic had been previously linked to several cases of blindness caused by attempts to use fat stem cells to treat macular degeneration.

The FDA also said it has taken "decisive action" to "prevent the use of a potentially dangerous and unproven treatment" offered by StemImmune Inc. of San Diego, Calif., and administered to patients at California Stem Cell Treatment Centers in Rancho Mirage and Beverly Hills, Calif.

As part of that action, the U.S. Marshals Service seized five vials of live vaccinia virus vaccine that is supposed to be reserved for people at high risk for smallpox but was being used as part of a stem-cell treatment for cancer, according to the FDA. "The unproven and potentially dangerous treatment was being injected intravenously and directly into patients' tumors," according to an FDA statement.

Smallpox essentially has been eradicated from the planet, but samples are kept in reserve in the U.S. and Russia, and vaccines are kept on hand as a result.

But Elliot Lander, medical director of the California Stem Cell Treatment Centers, denounced the FDA's actions in an interview with Shots.

"I think it's egregious," Lander says. "I think they made a mistake. I'm really baffled by this."

While his clinics do charge some patients for treatments that use stem cells derived from fat, Lander says, none of the cancer patients were charged and the treatments were administered as part of a carefully designed research study.

"Nobody was charged a single penny," Lander says. "We're just trying to move the field forward."

In a written statement, U.S. Stem Cell also defended its activities.

"The safety and health of our patients are our number one priority and the strict standards that we have in place follow the laws of the Food and Drug Administration," according to the statement.

"We have helped thousands of patients harness their own healing potential," the statement says. "It would be a mistake to limit these therapies from patients who need them when we are adhering to top industry standards."

But stem-cell researchers praised the FDA's actions.

"This is spectacular," says George Daley, dean of the Harvard Medical School and a leading stem-cell researcher. "This is the right thing to do."

Daley praised the FDA's promise to provide clear guidance soon for vetting legitimate stem-cell therapies while cracking down on "snake-oil salesmen" marketing unproven treatments.

Stem-cell research is "a major revolution in medicine. It's bound to ultimately deliver cures," Daley says. "But it's so early in the field," he adds. "Unfortunately, there are unscrupulous practitioners and clinics that are marketing therapies to patients, often at great expense, that haven't been proven to work and may be unsafe."

Others agreed.

"I see this is a major, positive step by the FDA," says Paul Knoepfler, a professor of cell biology at the University of of California, Davis, who has documented the proliferation of stem-cell clinics.

"I'm hoping that this signals a historic shift by the FDA to tackle the big problem of stem-cell clinics selling unapproved and sometimes dangerous stem cell "treatments" that may not be real treatments," Knoepfler says.

Kayhi grad represents Alaska at national camp

Class of 2017 Ketchikan High School graduate and Syracuse University freshman Cameron Edwards represented Alaska and Ketchikan at the 2017 National Youth Science Camp in Green Bank, West Virginia, this summer.

Edwards, who is studying to be an environmental engineer, joined 108 other delegates from 50 states and eight countries in the prestigious annual STEM (science, technology, engineering and mathematics) program. The event takes place in the Monongahela National Forest and provides learning opportunities through STEM presentations and outdoor activities.

We had some really cool presenters come, like the chief scientist for the International Space Station came, and one of the guys that helped pioneer stem cell research came and he talked about stem cells, Edwards said.

As a part of the event, the students travel to the nations capitol in Washington D.C., which is only five hours away from the camp. Each year, West Virginias senior member of the United States Senate honors the delegates with a luncheon attended by other members of the U.S. government.

We heard John McCain talk. We went to the senate building and theres a senate luncheon, and all of the senators are invited but it was the day that they were voting on the health care bill, so no one really came, Edwards said. Our nurse went and she was trying to find senators to come and talk and she found John McCain, he was walking out of the bathroom.

For nearly a month, the delegates attended lectures but also experienced the beauty of the West Virginia outdoors through backpacking, caving, mountain biking and rock climbing. Edwards said she particularly enjoyed the outdoor activities, but also learned a lot at the event.

At her favorite presentation, which she said wasnt necessarily scientific but rather related to the scientific community, she learned about teaching. Specifically, the lecturer talked about how to look at information and to question everything that they read, and how to change someones perspective, and what they believe to be true, without saying theyre wrong.

The camp is attended by graduating high school seniors with high achievements in STEM fields. The future STEM leaders are selected on merit, with the program provided to them at no cost, including travel, removing any financial barriers for attendance, according to a statement from the NYSC.

This is a unique experience for these amazing students and one that can impact them for the rest of their lives, John Giroir, director of the NYSC, said in a statement. Cameron is (an) excellent example of the kind of student who participates and goes on to significant achievements in the STEM fields.
