COLUMBIA, Mo. When a muscle is damaged, dormant adult stem cells called satellite cells are signaled to wake up and contribute to repairing the muscle. University of Missouri researchers recently found how even distant satellite cells could help with the repair, and are now learning how the stem cells travel within the tissue. This knowledge could ultimately help doctors more effectively treat muscle disorders such as muscular dystrophy, in which the muscle is easily damaged and the patients satellite cells have lost the ability to repair.

When your muscles are injured, they send out a mayday for satellite cells to come and fix them, and those cells know where to go to make more muscle cells, and eventually new muscle tissue, said D Cornelison, an associate professor of biological sciences in the College of Arts and Science and a researcher in the Bond Life Sciences Center. There is currently no effective satellite cell-based therapy for muscular dystrophy in humans. One problem with current treatments is that it requires 100 stem cell injections per square centimeter, and up to 4,000 injections in a single muscle for the patient, because the stem cells dont seem to be able to spread out very far. If we can learn how normal, healthy satellite cells are able to travel around in the muscles, clinical researchers might use that information to change how injected cells act and improve the efficiency of the treatment.

In a new study, researchers in Cornelisons lab used time-lapse microscopy to follow the movement of the satellite cells over narrow stripes of different proteins painted onto the glass slide. The researchers found that several versions of a protein called ephrin had the same effect on satellite cells: the cells that touch stripes made of ephrin immediately turn around and travel in a new direction.

The stem cell movement is similar to the way a person would act if asked to walk blindfolded down a hallway. They would feel for the walls, Cornelison said. Because the long, parallel muscle fibers carry these ephrin proteins on their surface, ephrin might be helping satellite cells move in a straighter line towards a distant mayday signal.

If researchers gave the satellite cells the signals to differentiate and form muscle fibers in culture, the group also found that they could use stripes of ephrins to get them to arrange themselves in parallel, the way muscle fibers always do in living beings, but have never been persuaded to do in a culture dish. This leads researchers to think that ephrins might actually be regulating several of the different steps that are needed to get from a population of stem cells spread out all over the muscle, to an organized and patterned new muscle fiber.

We are really excited about the potential of these findings to explain a lot of things that were puzzling about the way satellite cells behave in healthy muscle, compared to a muscular dystrophy patients own cells, or cells that have been injected therapeutically, Cornelison said. If were really lucky, we could find something that could make a difference in these kids lives, and thats what we want the most.

Michigan (Stem Cell) – what-when-how

A lot of work has been done in the state of Michigan on stem cells. The University of Michigan and prestigious hospitals such as the Henry Ford Hospital and many others have been working on human stromal cells and stem cells, finding out ways in which they can be used as potential life-saving cures and treatments. Much work has been carried out to improve the outcome after stroke, traumatic brain injury, and cancer.

The laws in the state of Michigan with regard to stem cell research are different from those in other states. Michigan law bans any research that destroys embryos for nontherapeutic purposes. Michigan law also bans the use of the somatic cell nuclear transfer procedure. Many of the states that have enacted legislation prohibiting human cloning have distinguished between reproductive and therapeutic cloning, but Michigan does not. Michigan, like South Dakota, forbids therapeutic cloning.

Michigan Citizens for Stem Cell Research and Cures is a nonprofit organization formed to educate the citizens of the state of Michigan, including public officials and policy makers, about the complex science, the biomedical potential, and the current policies affecting stem cell research in Michigan to promote informed decision making on this important issue.

In 2000 the researchers at the Henry Ford Health Sciences Center Department of Neurology tested a hypothesis and found that intracerebral grafting of a combination of bone marrow (BM) with brain-derived neurotrophic factor enhances differentiation of BM cells and significantly improves motor recovery. Because most of the active studies involve rats, before involving human beings, fresh BM was harvested from adult rats. It was anticipated that this may provide a powerful autoplastic therapy for human neurological injury and degenerative disorders.

In 2001 a significant step was taken by the Department of Neurosurgery at the Henry Ford Health Sciences Center. Rats were subjected to traumatic brain injury (TBI), and marrow stro-mal cells (MSCs) were injected into the tail vein 24 hours after TBI.

The rats were killed 15 days later. It was found that MSCs injected intravenously significantly reduced motor and neurological deficits. On the basis of this data, the researchers suggested that the intravenous administration of marrow stromal cells may be a promising therapeutic strategy that may be useful in treating TBI and that warrants further investigation.

The same year, researchers tested the hypothesis that intravenous infusion of bone marrow-derived MSCs enter the brain and reduce neurological functional deficits after stroke in rats. Significant recovery of somatosensory behavior was found. To test the efficacy of various delivery routes of stem cells, the researchers injected MSCs into the internal carotid artery of the adult rat after TBI. They came up with the suggestion that intra-arterial transplantation of MSCs along with intravenous and intracerebral transplantation is also a viable route for the administration of MSCs for the treatment of TBI, as MSCs infused intra-arterially after TBI survive and migrate into the brain.

As human umbilical cord blood cells (HUCBC) are rich in stem and progenitor cells, a team of researchers went ahead and tested whether intravenously infused HUCBC could enter the brain, survive, differentiate, and improve neurological functional recovery after stroke in rats. In addition, it was also tested whether ischemic brain tissue extract selectively induces chemotaxis of

HUCBC in vitro. Treatment with HUCBC significantly improved functional recovery, and significant HUCBC migration activity also was present. Therefore, HUCBC transplantation may provide a cell source to treat stroke.

The specific mechanisms by which introduced MSCs provide benefit remain to be elucidated. Various growth factors have been shown to mediate the repair and replacement of damaged tissue. Vascular endothelial growth factor (VEGF) is a growth factor responsible for growth of new vessels. It was confirmed that intravenous infusion of human bone marrow stromal cells promotes VEGF secretion, VEGF receptor 2 expression, and angiogenesis in the ischemic boundary zone of the host brain after stroke.

Water bucket challenge a game changer for marketing strategists, experts say.

Whether youve risen to the ALS Associations Ice Bucket Challenge with enthusiasm, or its oversaturation on social media has turned you into a wet blanket, local marketing experts said the craze has undeniably become a game changer for fundraising campaigns.

Sure, theres been some backlash. Some say its become more about vanity than charity. In the past two days, meanwhile, the Newark archdiocese, and Catholic dioceses in Ohio and Louisiana, have expressed concerns over the ALS Associations funding of research involving embryonic stem cells.

Nonetheless, not since breast cancers invention of the pink ribbon has a charity found such an explosively successful way to raise money and public awareness, according to industry observers. Other charities will have to do more than just come up with the next viral gimmick. Theyll have to invent something as innovative and groundbreaking, whatever it might be. Either that, or capitalize on the recent backlash against the craze.

But what have Facebook, Twitter and donating money ever been but a means for people to make themselves look good, marketing professionals ask. As long as it raises awareness and money, everybody wins.

How to explain why things go viral? In many ways, said industry observers, it isnt anything new. That bucket challenges surfaced on a grass-roots level and spread virally traces back to a business phenomenon as old as the hills: word of mouth, said Professor Ashwani Monga, chairman of the Marketing Department at Rutgers Business School in Newark.

"As we know, word of mouth matters most in any successful product or method. So in that sense, it has definitely worked," he said.

But why has the act of pouring cold water over our heads, for all the world to see, been such a digital draw? Again, observers said, the concept is steeped in timeless traditions born long before the advent of YouTube. Remember that concession at the school fundraising carnival, where you throw a ball and try to dunk your school principal in that big pool of cold water? Sound a little familiar?

"Theres no such thing as an original idea," said Anthony Torre, co-founder and chief marketing officer with Spitball Advertising in Red Bank. "Its a matter of transforming the idea in such a way that it becomes interesting again."

Both the carnival dunking booth and the ice bucket challenge share the same crucial selling point, he said: "Both are public forums in which we can make people look silly."

Except that now, rather than its being confined to the school principal, just about anyone who is anyone is willing to look silly if it means being seen on the Internet. And yes, while donating to a worthy cause. Locally, participants have run the gamut everyone from Bergen County Executive Kathleen Donovan, to Governor Christie, to various county freeholders, police officers and fire captains, some mayors, the Ridgewood High School boys soccer team, Audi Meadowlands General Manager Chris DeMarsico and 31 of his employees. And thats just to name a few.

Catholics asked to NOT donate to the ALS Association

DUBUQUE (KWWL) - The Archbishop of the Dubuque Diocese is asking Catholics participating in the ice bucket challenge to donate to the John Paul II Medical Research Institute in Iowa City instead of the ALS Association.

As of Wednesday, The ALS Association has received $94.3 million in donations, most of which are attributed to the recent ice bucket challenge.

Archbishop Michael Jackels has now released a statement urging Catholics participating to donate to another organization helping to fight ALS.

"Donating to the ALS Association is inconsistent with Catholic moral teaching as it promotes stem cell research that destroys embryonic human beings, Jackels said. Catholic individuals, parishes, schools, and other entities in the Archdiocese of Dubuque, if they participate in the Ice Bucket Challenge, are asked to send their donations to the John Paul II Medical Research Institute in Iowa City."

Other Catholic archbishops have asked the same of their congregations, and the ALS Association has responded to the church by saying, The ALS Association primarily funds adult stem cell research. Currently, The Association is funding one study using embryonic stem cells (ESC), and the stem cell line was established many years ago under ethical guidelines set by the National Institute of Neurological Disorders and Stroke (NINDS); this research is funded by one specific donor, who is committed to this area of research. In fact, donors may stipulate that their funds not be invested in this study or any stem cell project.

Boron Facilitates Stem Cell Growth and Development in Corn

Boron deficiency is one of the most widespread causes of reduced crop yield. Missouri and the eastern half of the United States are plagued by boron deficient soil and, often, corn and soybean farmers are required to supplement their soil with boron; however, little is known about the ways in which corn plants utilize the essential nutrient.

Now, researchers at the University of Missouri have found that boron plays an integral role in development and reproduction in corn plants. Scientists anticipate that understanding how corn uses the nutrient can help farmers make informed decisions in boron deficient areas and improve crop yields.

"Boron deficiency was already known to cause plants to stop growing, but our study showed that a lack of boron actually causes a problem in the meristems, or the stem cells of the plant," said Paula McSteen, associate professor in the Division of Biological Sciences and a researcher in the Bond Life Sciences Center at MU.

"That was completely unknown before. Through a series of experiments involving scientists from several disciplines at MU, we were able to piece together the puzzle and reach a new conclusion."

Meristems comprise the growing points for each plant, and every organ in the plant is developed from these specialized stem cells. Insufficient boron causes these growing points to disintegrate, affecting corn tassels and kernels adversely. When tassels are stunted, crop yields are reduced, McSteen said.

The research evaluated a group of plants stunted by its ability to grow tassels. Kim Phillips, a graduate student in McSteen's lab, mapped the corn plant's genome and found that a genetic mutation stunted tassel growth because it was unable to transport boron across the plant membranes, inhibiting further growth in the plants.

Amanda Durbak, a post-doctoral fellow in the College of Arts and Science at MU, also helped prove boron's usefulness to meristems. She treated two groups of tassel-less corn, one with a boron fertilizer and the other with only water. The group that was treated with boron grew normally, while the group treated with water withered.

Further testing revealed that, at the cellular level, the affected plants' meristems had altered pectin which is strengthened with boron and stabilizes the plant cell. Without the pectin, plant meristems disintegrate.

"By using various techniques and expertise at MU, including genomics, translational experiments with frog eggs, research in the field, cellular testing, and evaluations at the MU Research Reactor Analytical Chemistry facility and at MU Plant and Soil Analysis Facility, the study team drew conclusions that will help corn producers make informed decisions about raising crops in boron deficient zones," McSteen said.
